Overview

Neovascular endothelial cell membranes and associated macromolecules refer to the specialized surface structures of newly formed blood vessels, particularly those found within the tumor microenvironment or in vasoproliferative diseases (Thorpe, P. These membranes are characterized by the expression of specific markers that are absent or sequestered in mature, quiescent vasculature, such as anionic phospholipids like phosphatidylserine and various adhesion molecules (He, J., et al., 2009, Clinical Cancer Research). Biologically, these components facilitate the rapid proliferation, migration, and structural integrity of angiogenic vessels required for disease progression (Tozer, G. M., et al., 2005, Nature Reviews Cancer). In oncology, these macromolecules serve as docking sites for vascular targeting agents (VTAs) and vascular disrupting agents (VDAs) designed to selectively destroy tumor blood supply (Siemann, D. W., 2011, Expert Opinion on Investigational Drugs). For example, drugs like bavituximab target exposed phosphatidylserine to induce immune-mediated vessel destruction, while others target the cytoskeleton to cause rapid vascular collapse (Gerber, D. E., et al., 2011, Clinical Cancer Research). Beyond cancer, these targets are relevant in ophthalmology for treating conditions like age-related macular degeneration where pathological neovascularization occurs (Miller, J. W., 2005, American Journal of Ophthalmology). Targeting these structures offers a way to bypass the challenges of drug delivery to interstitial tumor cells by focusing on the accessible luminal surface of the blood vessels (Thorpe, P. E., 2004, Clinical Cancer Research).
