Nerve Growth Factor (NGF) is a well-characterized neurotrophic factor and neuropeptide, primarily known for its critical role in the growth, maintenance, proliferation, and survival of certain neurons. It is essential for the development and survival of sympathetic and sensory neurons and has roles beyond neural development, including immune function, stress response, and wound healing. Dysregulation is implicated in chronic pain.
NGF exerts its effects by binding to two main types of cell surface receptors: TrkA (NTRK1) and p75 Neurotrophin Receptor (p75NTR). The binding to TrkA promotes neuron growth and blocks apoptosis. Binding to p75NTR can lead to either cell survival or programmed cell death depending on co-receptor interactions.
