A nerve growth factor (NGF) modulator is any compound or agent that alters the activity of NGF, a neurotrophic protein essential for the growth, maintenance, proliferation, and survival of certain neurons. NGF modulators can either enhance or inhibit NGF signaling and are of significant interest in neuroscience and therapeutic development due to their roles in neuronal health, pain modulation, inflammation, and disease states. NGF interacts with its receptors TrkA (high-affinity) and p75NTR (low-affinity) to mediate these effects. Small endogenous ligands (e.g., ATP) have been shown to bind directly to NGF or its receptors to alter downstream signaling pathways. Dysregulation is implicated in chronic pain syndromes, and overactivity may contribute to cancer progression via inflammation modulation. Clinical trials have explored topical administration routes such as eye drops with good tolerability profiles so far. Both agonists/enhancers (for regeneration) and antagonists/inhibitors (for analgesia) are under investigation.
