The nerve growth factor (NGF) pathway is a critical neurotrophic signaling cascade involved in the regulation of growth, maintenance, proliferation, and survival of neurons—especially sympathetic and sensory neurons. NGF exerts its effects by binding to specific cell surface receptors: TrkA (high-affinity receptor for NGF), p75NTR (low-affinity pan-neurotrophin receptor). Activation initiates several intracellular pathways including Ras/MAPK (ERK), PI3K/Akt, PLCγ/Ca²⁺ pathways. Therapeutically, targeting the NGF pathway has been explored for treating neuropathies and chronic pain conditions due to its role in nociception modulation.
