p75NTR is a type I transmembrane glycoprotein receptor belonging to the tumor necrosis factor receptor (TNFR) superfamily. It acts as a pan-neurotrophin receptor, binding all neurotrophins and their proforms. p75NTR plays crucial roles in regulating cell death and survival, neuronal development, tissue remodeling, energy expenditure, hypoxic response, scar formation, and neurodegeneration. It's a key regulator of cell fate, influencing survival, apoptosis, neurite outgrowth and synaptic plasticity. The outcome of p75NTR activation depends on the specific ligand bound (mature vs. proneurotrophins), presence of co-receptors such as TrkA/B/C or sortilin, and the cellular context.
