Nescient helix-loop-helix 2 (NHLH2) is a transcription factor of the basic helix-loop-helix (bHLH) family, specifically of class A, that participates in the regulation of gene expression by binding to E-box motifs. NHLH2 is crucial in the development and function of the central and peripheral nervous system, particularly in neuroendocrine regulation and reproductive behavior. It is highly expressed in neurons involved in the hypothalamic-pituitary-gonadal axis, affecting body weight control, energy expenditure, and mating behavior. It acts as a transcriptional activator of genes such as NDN, PCSK1/3, MC4R, and KISS1, and regulates various neuropeptides influencing adult body weight and fertility. NHLH2 expression is controlled by hormonal signals (notably leptin and possibly estrogen), and it undergoes various post-translational modifications that modulate its activity. Loss of NHLH2 function in mice causes adult-onset obesity and infertility, and genetic variants in humans have been implicated in similar phenotypes. NHLH2 is not a classical therapeutic target (such as a druggable receptor or enzyme); it is primarily of interest as a transcriptional regulator and candidate risk gene in certain syndromes and metabolic conditions. Note: There are no known drugs directly targeting NHLH2 as of current knowledge, and its roles are mainly in genetic and developmental regulation rather than as a conventional pharmacological target.
