Overview

Netrin-G1 is a member of the netrin protein family, structurally distinct from classical secreted netrins due to its glycosyl phosphatidylinositol (GPI) membrane anchoring. There are at least six isoforms, mostly restricted to the central nervous system. Unlike classical netrins, netrin-G1 has unique C-terminal hydrophobic regions that facilitate its membrane association, and it does not show detectable affinity for standard netrin receptors. Netrin-G1 acts in axon guidance, synaptic specificity, and the formation of neural circuits. It is expressed in specific CNS neurons (notably thalamic relay neurons) and is crucial for proper synapse formation, dendritic patterning, and mechanosensory end organ assembly. Its ligand, NGL-1, is a postsynaptic adhesion protein facilitating trans-synaptic signaling. Mutations affecting netrin-G1 or its ligand NGL-1 are implicated in several neurodevelopmental pathologies, suggesting netrin-G1's critical role in the establishment and maintenance of functional neural networks[1][2][3].
