Overview

Netrin-G2 (NTNG2) is a vertebrate-specific, membrane-anchored axon guidance and synaptic cell adhesion molecule that plays a fundamental role in forming, maintaining, and diversifying neuronal circuits in the central nervous system[1][3][2]. Its extracellular domain mediates specific binding to its postsynaptic receptor NGL2, contributing to synaptic network organization and NMDA receptor-mediated signaling. NTNG2 is crucial for proper brain development and higher cognitive functions, with marked enrichment in regions such as the temporal lobe, claustrum, and insular cortex. Alterations in NTNG2 expression or pathogenic mutations have been implicated in diverse neurodevelopmental disorders (including intellectual disability and autism), epilepsy, and certain psychiatric disorders[3][1]. NTNG2 is also a prognostic biomarker in some cancers, but there are no known drugs or targeted therapeutics for this molecule as of now[4][5].
