Overview

Neudesin neurotrophic factor (NENF) is a secreted neurotrophic protein predominantly expressed in the central nervous system, with important roles in neuronal differentiation, survival, neurogenesis, and regulation of food intake. It belongs to the MAPR family and contains a cytochrome b5-like heme/steroid-binding domain essential for its activity. Neudesin activates MAPK and PI3K signaling pathways, possibly through a G protein-coupled receptor, although its specific receptor has not been identified. It exerts anorexigenic effects in the hypothalamus, is involved in maintaining hippocampal anxiety circuitry, and is expressed in various tissues, including adipose. Neudesin is overexpressed in several human cancers, where it may have oncogenic functions. Despite its multi-functional roles, no drugs are currently known to target neudesin directly, and the safety of manipulating its activity remains theoretical.

Mechanism of action

No drugs are known to target neudesin directly. Endogenous neudesin acts via activation of MAPK and PI3K pathways, possibly through interaction with (yet unidentified) G protein-coupled receptors or heme-binding.

Biological functions

Neuronal differentiationNeuronal survivalNeurogenesisCell proliferation (primarily in neural progenitors)Apoptosis regulation (decreases neuronal apoptosis)Regulation of food intake (anorexigenic factor)Energy metabolism (affecting adipose tissue and systemic metabolism)Tumorigenesis (overexpressed in cancers, stimulating proliferation)Anxiety circuitry maintenance in hippocampus
04

Disease associations

Neurodegenerative disease (potential neuroprotection, role in anxiety circuitry)Obesity (affects hypothalamic food intake regulation and adipose metabolism)Cancer (overexpression or induced by DNA hypomethylation stimulates tumorigenesis)Other (possible involvement in neural developmental disorders, energy metabolism abnormalities)
05

Safety considerations

As NENF/neudesin is neurotrophic and affects multiple systems (CNS, metabolism), modulation may risk neurodevelopmental impacts, childhood development, appetite regulation, mood/anxiety, and oncogenicity if used therapeuticallyLack of a known specific receptor or approved drugs presents a challenge for specificity and safety as a drug target
06

Interacting drugs

No drugs directly targeting neudesin/NENF are currently known in clinical use or published literature
07

Biomarkers

Expression or overexpression of neudesin/NENF may serve as a biomarker in some cancersMay be investigated as a biomarker for obesity susceptibility or metabolic abnormalitiesNo validated biomarkers for clinical patient selection or efficacy monitoring
