Overview

Brain networks represent the complex, hierarchical organization of structural and functional connections between distinct brain regions and neuronal populations. Rather than acting as a single molecular entity, these networks integrate signaling across multiple neurotransmitter systems and cellular layers to facilitate complex behaviors and cognitive functions (Bullmore & Sporns, 2009, Nature Reviews Neuroscience). Pathological disruptions in network topology, often characterized as 'connectopathies,' are central to the manifestation of disorders such as schizophrenia, Alzheimer’s disease, and major depressive disorder (Fornito et al., 2015, Nature Reviews Neuroscience). While traditional pharmacology indirectly influences these networks by modulating synaptic weights, modern therapeutic approaches such as Transcranial Magnetic Stimulation (TMS) and Deep Brain Stimulation (DBS) target these circuits directly to rectify aberrant firing patterns. Consequently, the brain network is a macro-scale system target that necessitates an understanding of both molecular signaling and large-scale topological dynamics for effective clinical intervention (NIH National Institute of Mental Health, 2023).
