The NEDD8-activating enzyme (NAE) is a crucial E1 enzyme in the neddylation pathway, responsible for activating the ubiquitin-like protein NEDD8. This activation is essential for the post-translational modification of target proteins, primarily cullin family members, which are key components of cullin-RING ubiquitin ligases (CRLs). The neddylation process regulates protein degradation and turnover, impacting cell cycle progression, DNA damage response, stress signaling, and other vital cellular processes. Aberrant neddylation contributes to tumorigenesis and viral replication, making NAE a valid therapeutic target.
