Overview

Neural precursor cell expressed, developmentally down-regulated 9 (NEDD9) is a multi-domain, non-catalytic scaffolding protein belonging to the CAS family. It primarily assembles and regulates signaling complexes relevant to cell migration, adhesion, invasion, chemotaxis, cell cycle progression, and apoptosis. NEDD9 expression is tightly regulated during development, being most prominent in neural precursor cells and downregulated in differentiating tissues. In cancers, NEDD9 is frequently upregulated and hyperphosphorylated, which amplifies oncogenic signaling and supports tumor growth, metastasis, and chemotherapy resistance. Although not itself a direct oncogene, its dysregulation is associated with poor clinical outcomes in several malignancies, and it serves as both a functional target and a biomarker for disease progression and therapy response. NEDD9 also plays roles in non-malignant processes including brain development, neuronal differentiation, and cell adhesion in the immune system
