Overview

NPDC1 is a protein expressed primarily in the brain and lung, correlating with the regulation of cellular proliferation and differentiation. Its expression is linked to withdrawal from the cell cycle and entry into differentiation in neural cells. NPDC1 interacts with E2F1 transcription factor, cyclin D1, and Cdk2, modulating cell cycle progression. It regulates retinoic acid-mediated transcription and is targeted for degradation by the ubiquitin/proteasome system, specifically via a PEST motif in its carboxyl terminus. Phosphorylation regulates its stability and degradation rate. NPDC1 has been mapped to human chromosome 9q34.3. Its primary documented function is to suppress oncogenic transformation and down-regulate neural cell proliferation. NPDC1 is well-characterized as a regulatory protein in neural development rather than as a direct drug target. No interacting drugs, biomarker status, or safety challenges are documented in the available literature.
