Overview

Neuralized E3 ubiquitin protein ligase 1B (NEURL1B) is an enzyme in the neuralized family of E3 ubiquitin ligases, playing a regulatory role in the ubiquitin-proteasome pathway by mediating the ubiquitination of specific substrate proteins such as Notch ligands. It is involved in the modulation of Notch signaling, influencing cellular differentiation, particularly during neurodevelopment. NEURL1B is localized to the actin cytoskeleton and cytosol. It is predicted to contribute to ubiquitin-dependent endocytosis but distinct from related enzymes (e.g., NEURL1), may have unique or only partially overlapping substrates and roles. Genetic associations suggest potential roles in congenital and developmental disorders, with limited direct disease linkages known to date[3][5][6].
