Overview

Neuralized E3 ubiquitin protein ligase 2 (NEURL2) is a cytosolic E3 ubiquitin ligase enzyme that functions as an adaptor component in SCF-like ECS (Elongin BC-CUL2/5-SOCS-box protein) complexes, targeting specific substrate proteins for ubiquitination and subsequent proteasomal degradation. NEURL2 plays a critical role in striated muscle, regulating myofibril organization and specifically mediates the degradation of beta-catenin during myogenesis. In stem cell biology, NEURL2 acts as an asymmetric division gene and is directly repressed by the EMT transcription factor Twist2, with implications in cancer stem cell self-renewal and prognosis for lung adenocarcinoma. NEURL2 is therefore considered both a key molecular regulator in muscle and stem cell biology and a potential biomarker for cancer prognosis. Alternative splicing leads to multiple transcript variants. Disease associations include muscular dystrophy and galactosialidosis. No known drugs target NEURL2 directly, and no safety concerns are described in current research.
