Overview

Neuralized E3 ubiquitin protein ligase 4 (NEURL4) is a centriolar protein belonging to the Neuralized family, characterized by E3 ubiquitin ligase activity. It primarily localizes to the daughter centriole and interacts with CP110, promoting its ubiquitination and subsequent proteasomal degradation. This post-translational modification of CP110 by NEURL4 is essential for the maintenance of centriolar homeostasis and prevention of ectopic microtubule-organizing centers, thereby ensuring normal cell division and centrosome function. Loss or depletion of NEURL4 leads to accumulation of CP110, formation of aberrant centrosome structures, and mitotic defects, which are processes relevant to cellular genomic stability but not directly targeted by current therapeutics[1][2][7].
