Overview

Neuregulin 1 is a multifunctional glycoprotein belonging to the EGF family that signals primarily through ErbB receptor tyrosine kinases (especially ErbB2, ErbB3, and ErbB4)[1][2][3][5]. It exists in numerous isoforms due to extensive alternative splicing, and plays essential roles in the development, maintenance, and function of the nervous and cardiovascular systems, as well as glial, epithelial, and other cell types[1][2]. NRG1's biological effect is mediated by direct activation of ERBB receptors, triggering downstream signaling pathways including Ras-MAPK and PI3K–Akt, to modulate cellular survival, growth, differentiation, synaptic plasticity, and neurotransmission[2][3][5]. Dysregulation of Neuregulin 1 is associated with multiple cancers (sometimes acting as a tumor suppressor, but also involved in oncogenic fusion events), schizophrenia, depression, and neurodegenerative diseases[1][3][5][6]. It is widely studied as a therapeutic target, especially in oncology and neurology[3].

Mechanism of action

Agonism/antagonism at ERBB receptors: Drugs may block or modulate downstream NRG1 pathways by inhibiting ErbB activity in tumors expressing NRG1 fusion proteins. Pathway inhibition: e.g., inhibition of Ras-MAPK and PI3K-Akt signaling cascades. Indirect modulation of synaptic plasticity, myelination, Schwann cell survival, neuronal growth via ErbB receptor blockade or activation
