Overview

Neurexophilin-4 (NXPH4) is a secreted glycoprotein member of the neurexophilin family that functions as a selective ligand for alpha-neurexins and interacts with GABA_A receptors, predominantly within certain brain circuits and at synaptic regions, especially in the cerebellum[2][3]. Its biological activity appears to modulate specific synaptic functions, facilitating inhibitory neurotransmission and contributing to motor coordination, feeding, emotion, and balance[2]. High expression of NXPH4 has been associated with metabolic reprogramming, immune infiltration, and poor prognosis in multiple cancers, especially colon adenocarcinoma, suggesting a potential oncogenic role and biomarker utility for cancer progression and prognosis[1]. NXPH4 is not currently targeted by any known drugs, nor is it an established enzyme, receptor, or transporter; rather, it supports synaptic function through protein-protein interactions in the nervous system[2][3]. Loss of NXPH4 impairs cerebellar inhibitory synaptic transmission and may therefore contribute to neurological dysfunction, though direct disease associations outside cancer are not well-characterized.
