Neuroblastoma highly expressed DDX5 stabilizing lncRNA 1 (NHEG1) is a long non-coding RNA prominently upregulated in neuroblastoma tissues, where it influences tumor progression and aggressiveness. It functions by binding to and stabilizing the RNA helicase DDX5, thereby inhibiting its endogenous ubiquitination and proteasomal degradation. This stabilization leads to increased β-catenin activity, promoting cell proliferation and aggressive tumor behavior. NHEG1 also acts as a competitive endogenous RNA for miR-665, which results in derepression of HMGB1, a protein associated with stemness and invasiveness in neuroblastoma. High expression of NHEG1 correlates with poor outcomes and could serve as a prognostic biomarker for neuroblastoma. There are currently no known drugs or drug mechanisms directly targeting NHEG1, nor are there notable safety concerns reported for therapeutic targeting of this molecule.
