Overview

Neuroblastoma suppressor of tumorigenicity 1 (NBL1) is the founding member of the DAN family of bone morphogenetic protein (BMP) antagonists. It is a secreted protein containing a CTCK/cystine knot-like motif and acts in the extracellular space to bind and sequester BMP ligands, thereby preventing their interaction with BMP receptors. NBL1 regulates various developmental processes (including neurogenesis and osteogenesis), cell cycle progression, and immune cell migration. Dysregulation or aberrant expression of NBL1 is associated with a variety of diseases including a range of cancers (acting as a tumor suppressor), renal diseases (where increased serum levels mark poor prognosis), and disorders of development resulting from imbalanced BMP signaling
