Overview

Neurochondrin (NCDN) is a highly conserved 79 kDa adaptor protein encoded by the NCDN gene, with prominent expression in the nervous system. It plays essential roles in neuronal morphology, synaptic plasticity, and normal brain development and homeostasis. It regulates cell signalling chiefly by interacting with a broad range of G protein-coupled receptors (GPCRs), influencing their trafficking and signalling. Neurochondrin also interacts with molecular regulators such as the Rac guanine-nucleotide exchange factor P-Rex1 and protein kinase A, affecting downstream signalling pathways including ERK, Ca^2+, and small GTPases like Rac. The protein lacks known catalytic activity or defined protein domains, featuring a largely α-helical, leucine-rich structure thought important for protein–protein interactions. Dysregulation and genetic variants of NCDN/Norbin are implicated in several neurological disorders, including neurodevelopmental and neuropsychiatric conditions such as epilepsy, intellectual disability, and changes associated with schizophrenia. Recently, NCDN has also been shown to suppress neutrophil-driven innate immunity, impacting antimicrobial responses.
