Overview

Neuroepithelial cell-transforming gene 1 protein (NET1) is a RhoA-specific guanine nucleotide exchange factor (GEF) that catalyzes the activation of the small GTPase RhoA by promoting GDP–GTP exchange[1][3][4]. NET1 orchestrates cytoskeletal dynamics, cell adhesion, and motility by stimulating actin stress fiber and focal adhesion formation, which modulates extracellular matrix degradation and promotes invasive cellular behavior[1][3]. Its activity is controlled by subcellular localization, phosphorylation, and protein–protein interactions, with nuclear–cytoplasmic shuttling playing a key role in its regulation[1]. Aberrant or elevated NET1 expression is strongly linked to tumor progression, invasion, metastasis, and chemoresistance in several human cancers, and it may serve as both a prognostic biomarker and an emerging therapeutic target[1][3][2]. While targeting the NET1–RhoA interaction has shown experimental promise as an anticancer strategy[2], no clinically approved drugs against NET1 currently exist.

Mechanism of action

Inhibitors or peptides that disrupt NET1–RhoA binding block guanine nucleotide exchange on RhoA, inhibiting downstream RhoA-mediated signaling involved in cancer cell invasion and motility[2].

Safety considerations

As a central signaling regulator, systemic inhibition of NET1 could interfere with normal cytoskeletal function, cell migration, adhesion, DNA repair and may pose risks of impaired wound healing or tissue repair, as well as immune and cell viability effects. No clinical safety data is available.

No approved or clinically validated drugs are currently known to target NET1 directly; research peptides have been shown to disrupt the NET1–RhoA interaction in studies[2].

Elevated NET1 expression levels have been identified as a prognostic marker in various cancers (effect on tumor cell proliferation, invasion, metastasis, and poor clinical outcomes)[1].
