Neurogenic differentiation factor 6 (NEUROD6) is a basic helix-loop-helix (bHLH) transcription factor that plays a critical role in the development and maintenance of the mammalian nervous system. It is highly expressed during embryonic and early postnatal brain development, particularly in pyramidal neurons of the cortex and hippocampus. NEUROD6 regulates neuronal differentiation, axon guidance, mitochondrial biogenesis, and antioxidant defense, promoting neuronal survival under conditions of trophic factor withdrawal and oxidative stress. Loss of NeuroD6 function in mice leads to defects in neuronal differentiation and axon pathfinding, with implications for neurodevelopmental pathologies and possibly neurodegenerative conditions. While it is linked to several neurological diseases as a genetic locus, NEUROD6 itself is not a direct therapeutic target for drugs, nor is it a receptor, enzyme, or transporter, but rather a key developmental transcription factor involved in broad regulatory networks in neurons.
