Overview

Notch receptor 2 (NOTCH2) is a type I transmembrane protein and a member of the Notch family of receptors. It plays a critical role in cell fate determination during embryonic development and adult tissue homeostasis by mediating intercellular signaling. Upon ligand binding, NOTCH2 undergoes proteolytic cleavages, releasing the NICD which regulates gene expression. Dysregulation is implicated in diseases like Alagille syndrome, Hajdu–Cheney syndrome, and various cancers.

Mechanism of action

Ligand binding induces proteolytic cleavage, releasing the NICD which translocates to the nucleus and activates target genes through interaction with CSL and Mastermind-like proteins.
