Overview

Neurogenin-3 (NEUROG3) is a basic helix-loop-helix transcription factor essential for the development of endocrine lineages in the pancreas and intestine. NEUROG3 governs the specification and differentiation of pancreatic islet cells (including insulin-producing beta cells) and mature enteroendocrine cells of the intestine. Mutations cause congenital absence of enteroendocrine cells, leading to malabsorptive diarrhea and sometimes neonatal or early-onset diabetes. NEUROG3 acts mainly by binding to E-box sequences in target gene promoters—often as a heterodimer with E proteins like E47—regulating gene transcription involved in endocrine cell fate. Its expression and activity are tightly controlled at transcriptional and post-translational levels, with rapid turnover via the ubiquitin-proteasome system and regulation by phosphorylation and deubiquitination[1][2][3].
