Overview

Alpha-synuclein-induced neuroinflammation is a key pathogenic mechanism in synucleinopathies. Misfolded and aggregated alpha-synuclein activates microglia via TLR2/TLR4, leading to NF-κB activation and NLRP3 inflammasome assembly. This results in the release of pro-inflammatory cytokines (IL-1β, IL-18, TNFα) that contribute to neuronal damage and death. Peripheral immune cell infiltration and dysregulation of microRNAs like miR155 further exacerbate the inflammatory response. Therapeutic strategies aim to reduce alpha-synuclein aggregation, modulate immune signaling pathways, and protect neurons from inflammation-mediated injury.
