Overview

Neurokinin B (NKB) is a decapeptide member of the tachykinin family encoded by the TAC3 gene in humans. It acts primarily as an endogenous agonist at the Neurokinin 3 receptor (NK3R), which belongs to the G protein-coupled receptor superfamily. NKB plays critical roles in neuronal excitation, smooth muscle contraction, vasodilation, and especially regulation of gonadotropin-releasing hormone secretion—a key process for normal reproductive function. Mutations affecting either NKB or its receptor can result in hypogonadotropic hypogonadism due to impaired GnRH release. Elevated levels have been implicated in pre-eclampsia during pregnancy. While not itself targeted by drugs, modulation of its signaling through NK3R antagonists has been explored therapeutically for conditions such as menopausal hot flashes and psychiatric disorders.
