Overview

Neuroligin-1 (NLGN1) is a postsynaptic cell adhesion protein encoded by the NLGN1 gene, belonging to the neuroligin family. It predominantly localizes to the postsynaptic membranes of excitatory (glutamatergic) synapses, where it acts as a ligand for presynaptic β-neurexins. Through trans-synaptic interactions, NLGN1 plays a crucial role in the formation, differentiation, and modulation of synapses in the central nervous system. Regulation of NLGN1, including its cleavage and phosphorylation by receptor tyrosine kinases such as TrkB, affects synaptic structure and functional plasticity. Aberrations in NLGN1 levels or sequence are linked to multiple neurodevelopmental, neuropsychiatric, and neurodegenerative conditions, making it a significant molecule in the study of brain disorders and synaptic biology[1][2][3][4].
