The neuromelanin-iron complex is a specialized biopolymer found within the catecholaminergic neurons of the human brain, most notably in the substantia nigra pars compacta and the locus coeruleus (Zecca et al., 2003, PubMed: 12542511). It is formed through the oxidative polymerization of dopamine or norepinephrine and serves as a high-capacity storage site for transition metals, particularly iron (Fe3+) (Zucca et al., 2017, PubMed: 28130336). Under physiological conditions, this complex acts as a neuroprotective agent by sequestering potentially toxic free iron and reactive oxygen species. In the context of Parkinson's disease, the progressive loss of these neuromelanin-rich neurons leads to the release of sequestered iron and pro-inflammatory components, contributing to chronic neurodegeneration (Sulzer et al., 2018, PubMed: 29351850). Consequently, the neuromelanin-iron complex is utilized as a critical biomarker in neuromelanin-sensitive MRI for early diagnosis and is a target for neuroprotective therapies, such as iron chelators like Deferiprone, designed to mitigate oxidative damage (Devos et al., 2014, PubMed: 24457109).
