Overview

Neuronal guanine nucleotide exchange factor (NGEF), also known as Ephexin-1 or ARHGEF27, is a Dbl family guanine nucleotide exchange factor predominantly expressed in the central nervous system. It acts downstream of Eph tyrosine kinase receptors, linking Eph/ephrin signals to RhoA, Rac, and Cdc42 GTPases, and is crucial for axon guidance, synaptic homeostasis, and cytoskeletal remodeling. NGEF is an enzymatic signaling hub, modulating actin dynamics in response to neuronal and developmental cues. Dysregulation of NGEF/Ephexin-1 function is implicated in various cancer types and neurological disorders, making it a relevant target for research into neurodevelopmental diseases and cancer therapies
