Sporadic kidney cancer gene 1 (SKCG-1) is a candidate tumor suppressor gene located on chromosome 11q23.2, first identified in the context of sporadic Wilms tumors and breast cancer. It encodes a predicted 124-amino acid transmembrane protein that plays a critical role in regulating cell growth and inhibiting proliferation in kidney and breast tissues. Research has shown that genomic loss or significantly reduced expression of SKCG-1 is common in approximately 38% of sporadic Wilms tumors, particularly those with high proliferative indices. Experimental silencing of the gene via siRNA in human embryonic kidney epithelial cells leads to a marked increase in cell growth, further supporting its role as a growth regulator. While it is considered a potential therapeutic target for restoring growth control in renal and breast malignancies, no specific drugs currently target SKCG-1 in clinical practice. The gene's identification provides valuable insights into the molecular pathogenesis of pediatric kidney cancers and may serve as a biomarker for tumor progression. Its predicted structure as a transmembrane protein suggests it may be involved in signal transduction pathways that control the cell cycle. Despite its potential, the gene remains relatively understudied compared to other Wilms tumor genes like WT1.
