Overview

Because this term combines a structural component (the membrane) with a diverse group of receptor families, it is generally treated as a functional system rather than a single, specific therapeutic target in drug discovery.

Mechanism of action

Drugs targeting this system typically act as agonists or positive allosteric modulators of inhibitory receptors, such as GABA-A or GABA-B, to increase chloride influx or potassium efflux. This results in the hyperpolarization of the neuronal membrane, thereby reducing the probability of action potential firing and dampening overall neuronal excitability (Olsen & Sieghart, 2008; Sigel & Steinmann, 2012).
