Overview

The neuronal nicotinic acetylcholine receptor alpha-4-alpha-6-beta-2 is a heteromeric ligand-gated ion channel subtype primarily expressed in the central nervous system (UniProt P43681, Q15825, P17676). It is notably localized in the dopaminergic neurons of the ventral tegmental area and the substantia nigra, where it modulates the release of dopamine in the striatum (PMID: 17030626). This specific subunit configuration makes the receptor a key mediator of the rewarding properties of nicotine and a primary target for smoking cessation therapies. In addition to its role in addiction, the receptor is involved in the regulation of locomotor activity and has been linked to the pathology of Parkinson's disease. Therapeutic strategies often utilize partial agonists to provide a controlled level of receptor activation while blocking the more intense stimulation caused by nicotine. Selective targeting of this receptor is a major goal in drug development to avoid side effects associated with the activation of peripheral nicotinic receptors (PMID: 21841201). Research indicates that these receptors may also serve as potential targets for treating cognitive impairments and certain mood disorders.
