The neuronal nicotinic receptor alpha-7 subtype (α7 nAChR) is a homopentameric ligand-gated ion channel highly permeable to calcium ions (Ca²⁺), in addition to sodium (Na⁺) and potassium (K⁺). It is widely expressed in the central nervous system and peripheral tissues. It mediates fast synaptic transmission, modulates neurotransmitter release, and is involved in cognitive processes, neurogenesis and inflammation regulation. It has been implicated in neuroprotection, cognitive disorders like schizophrenia and Alzheimer's disease, and cancer progression.
Activation leads to rapid desensitization but also supports neuronal survival after excitotoxic stimuli via Ca²⁺ dependent mechanisms; inhibits pro-inflammatory cytokine release from immune cells.
