Overview

Neuronal plasma membrane phospholipids are essential structural and functional components of the nervous system, primarily composed of phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, and phosphatidylinositol (PMID: 12070353). These lipids form the matrix of the neuronal bilayer, maintaining the fluidity and integrity necessary for the proper orientation and function of membrane-bound proteins such as ion channels, transporters, and receptors (PMID: 16456348). Beyond their structural role, they serve as precursors for bioactive signaling molecules like diacylglycerol and inositol triphosphate, and are vital for the processes of synaptic vesicle fusion and neurotransmitter release (PMID: 10479465). In neurodegenerative conditions like Alzheimer's and Parkinson's diseases, the composition and metabolism of these phospholipids are often significantly altered, leading to impaired synaptic transmission and increased neuronal vulnerability (PMID: 25933483). Therapeutic strategies targeting these phospholipids include the administration of lipid precursors or stabilizers, such as Citicoline or Docosahexaenoic acid (DHA), to restore membrane homeostasis and enhance cognitive function (PMID: 16456348).
