Neuronal protection markers represent a heterogeneous group of biological indicators used to evaluate the preservation of neuronal structure and function in response to pathological insults or therapeutic interventions. This term does not refer to a single molecular target but rather a functional category of proteins and molecules, such as Brain-derived neurotrophic factor (BDNF), Neurofilament light chain (NfL), and Neuron-specific enolase (NSE), which serve as proxies for neuronal viability (Gaetani et al., 2019, J Neurol Neurosurg Psychiatry). These markers are critical in the study of neurodegenerative diseases like Alzheimer's and Parkinson's, as well as acute injuries like stroke and traumatic brain injury, where they help quantify the extent of damage or the success of neuroprotective strategies (Papa et al., 2008, Acad Emerg Med). In drug development, these markers are utilized as pharmacodynamic endpoints to demonstrate that a drug candidate is effectively engaging with pathways that promote cell survival or prevent apoptosis (Huang & Reichardt, 2001, Annu Rev Neurosci). Because the term encompasses a wide array of molecules with diverse biological roles—ranging from structural components to growth factors—it is classified as a descriptive category rather than a specific therapeutic target. Consequently, while individual markers within this group may be targeted or monitored, the collective term itself is not a druggable entity.
