Overview

Neuropeptide receptors are a diverse group of cell surface proteins that primarily mediate the effects of neuropeptides—small protein-like molecules used by neurons to communicate with each other and with other cell types. These receptors are found throughout the nervous system and in various peripheral tissues, where they play key roles in modulating neural activity, physiological processes, and behavior. Most neuropeptide receptors belong to the G protein-coupled receptor (GPCR) superfamily, specifically characterized by seven transmembrane domains. Upon activation, these receptors initiate intracellular signaling cascades via G proteins. This leads to changes in second messenger levels (such as cAMP or IP3), ultimately modulating cellular responses over longer time scales compared to classical neurotransmitters. Neuropeptide signaling is typically slower but more modulatory than fast synaptic transmission.
