Overview

Neuropeptide Y receptor type 5 (NPY5R) is a G protein-coupled receptor (GPCR) that binds neuropeptide Y (NPY), peptide YY, and pancreatic polypeptide. It plays a role in appetite regulation, energy homeostasis, modulation of neuronal activity, cell growth signaling, and synaptic transmission. It has been implicated in epilepsy and certain cancers such as neuroblastoma, breast cancer and liver cancer.

Mechanism of action

Activation of presynaptic Y5 receptors induces a sustained increase in spontaneous GABA release from inhibitory neurons. Activation inhibits adenylate cyclase activity leading to decreased cAMP levels. Activation can also trigger ERK signaling pathways promoting cell proliferation.
