The neurotransmitter release machinery refers to the highly specialized molecular system responsible for the exocytosis of neurotransmitters from presynaptic neurons into the synaptic cleft, enabling communication between neurons or between neurons and other cell types. This process is fundamental to synaptic transmission and underlies all neural network activity. It involves vesicle docking & priming, calcium triggering, SNARE complex formation, membrane fusion, neurotransmitter release, and termination & recycling. Key components include SNAREs (syntaxin-1, SNAP25, VAMP/synaptobrevin), synaptotagmin, complexin, Munc18/Munc13, and RIM/RIM-BP. Disruption of this machinery leads to neurological disorders.
