NTRK1 encodes a transmembrane receptor tyrosine kinase that is the high-affinity receptor for nerve growth factor beta (NGFβ). Upon ligand binding, NTRK1 undergoes dimerization and autophosphorylation, activating downstream signaling pathways such as MAPK/ERK and PI3K/Akt, which are essential for neuronal differentiation, survival, and proliferation. NTRK1 plays a critical role in the development and function of sensory neurons. Aberrant NTRK1 signaling, particularly through gene fusions, contributes to oncogenesis in various cancers. Loss-of-function mutations cause Congenital Insensitivity to Pain with Anhidrosis.
