Overview

The neurotrophic receptor tyrosine kinase (TRK) family consists of TrkA, TrkB, and TrkC, which are high-affinity receptors for neurotrophins. These receptors play crucial roles in the development, function, and maintenance of the nervous system. Upon ligand binding, TRKs dimerize and autophosphorylate, activating downstream pathways like RAS/MAPK and PI3K/Akt, influencing neuronal survival, differentiation, and synaptic plasticity. Dysregulation or mutations involving NTRK genes, particularly gene fusions, can drive oncogenesis in various cancers. They interact with other molecules such as NCAMs further modulating neurite outgrowth.
