The neurotrophic support system encompasses the action of neurotrophic factors, such as NGF, BDNF, NT-3, and NT-4/5, which promote the growth, survival, differentiation, and maintenance of neurons. These factors signal through Trk receptors (TrkA, TrkB, TrkC) and the p75NTR receptor, influencing neuronal plasticity, synaptic efficacy, learning, memory formation, and regeneration after injury. Dysregulation of this system is implicated in various neurodegenerative and psychiatric disorders.
Activation of Trk receptors (TrkA, TrkB, TrkC) leads to downstream signaling cascades including MAPK and PI3K/Akt pathways, which promote neuronal survival, growth, and differentiation. Modulation of p75NTR can either enhance or antagonize Trk signaling.
