Overview

Neurotrophic tropomyosin receptor kinases (NTRKs) are a family of transmembrane receptor tyrosine kinases that play essential roles in the development, function, and survival of neurons. The NTRK gene family includes three main members: NTRK1, NTRK2, and NTRK3, which encode the TRKA, TRKB, and TRKC proteins respectively. Chromosomal rearrangements involving *NTRK* genes can result in fusion proteins that drive constitutive activation of the kinase domain, acting as oncogenic drivers across many cancer types. Selective inhibitors targeting these fusion proteins, such as larotrectinib and entrectinib, are approved therapies for solid tumors harboring confirmed pathogenic *NTRK* gene fusions.
