Overview

Neurotrophin-4 (NTF4) is a member of the neurotrophin family of growth factors responsible for the survival, development, and function of neurons in the central and peripheral nervous systems[1][2][8]. Unlike other neurotrophins, NTF4-deficient mice only show minor cellular changes and generally develop normally[1]. NTF4 signals mainly through the TrkB tyrosine kinase receptor, driving neuronal survival and differentiation, and may also promote ameloblast differentiation[2][3][8]. Pathological variants in the NTF4 gene are associated with certain types of glaucoma[1][6]. There are no currently approved drugs that specifically target NTF4, but modulation of its downstream pathways (especially via TrkB) remains a research interest for neuroprotection and neurodegenerative disease[2].
