Overview

Neutrophil cytoplasmic factor 2 (NCF2), also known as p67-phox, is a critical cytosolic subunit of the multi-protein NADPH oxidase (NOX2) complex found primarily in phagocytes (UniProt P19878). Upon activation, NCF2 translocates to the plasma membrane where it interacts with other subunits like p47-phox and cytochrome b-558 to initiate the production of superoxide anions, a process known as the respiratory burst (PubMed: 2333537). This mechanism is essential for the microbicidal activity of neutrophils and macrophages. Mutations in the NCF2 gene are a known cause of autosomal recessive chronic granulomatous disease (CGD), characterized by recurrent life-threatening bacterial and fungal infections (NIH: GARD). Beyond its role in host defense, overactivation of the NOX2 complex and NCF2-mediated ROS production are implicated in chronic inflammatory conditions and autoimmune diseases like systemic lupus erythematosus (PubMed: 29038200). Consequently, NCF2 and the NADPH oxidase complex are targets for therapeutic intervention aimed at modulating oxidative stress in inflammatory and cardiovascular disorders.
