Overview

Neutrophil cytosolic factor 2 (NCF2, p67-phox) is a regulatory and catalytic protein subunit of the multi-protein NADPH oxidase complex, predominantly expressed in phagocytes such as neutrophils. It is essential for assembling the active enzyme complex that generates reactive oxygen species (mainly superoxide) during the oxidative burst, a critical process for destroying pathogens and regulating immune cell signaling. Mutations in the NCF2 gene cause impaired ROS production leading to chronic granulomatous disease, characterized by recurrent infections and abnormal inflammation. Variations in NCF2 also contribute to autoimmunity (e.g., systemic lupus erythematosus) and are emerging as a biomarker of aggressive behavior in several cancers, including esophageal squamous cell carcinoma[1][2][3][4][6][7][8].
