Overview

Nexilin F-actin binding protein (NEXN) is a cytoskeletal protein that binds filamentous actin and is essential for maintaining the structural integrity of Z-discs in cardiomyocytes and skeletal muscle. It is crucial for mechanical stability during muscle contraction and tissue remodeling, particularly in the heart and vascular smooth muscle. NEXN mutations impair myofibrillar structure, leading to dilated and hypertrophic cardiomyopathies, and may be implicated in vascular remodeling processes. No direct drugs target NEXN, but pathogenic variants serve as important genetic biomarkers for inherited cardiomyopathies
