Overview

NF-kappa-B inhibitor beta is a cytoplasmic regulatory protein encoded by the NFKBIB gene that belongs to the NF-kappa-B inhibitor family[1][3][4][5][8]. Its primary role is to suppress the activity of the NF-kappa-B transcription factor complex by sequestering it in the cytoplasm, preventing translocation to the nucleus and subsequent DNA binding. Upon cellular stimulation, NFKBIB is phosphorylated by I-kappa-B kinases, leading to its degradation via the ubiquitin-proteasome pathway and freeing NF-kappa-B to initiate gene transcription, crucial for immune and inflammatory responses. Mechanism of action

Drugs targeting the NF-kappa-B pathway may act by preventing the phosphorylation or degradation of NFKBIB, stabilizing its inhibitory complex with NF-kappa-B and keeping the transcription factor inactive in the cytoplasm.
