Overview

NF-kappa-B inhibitor epsilon (NFKBIE) is a cytoplasmic protein that functions as an inhibitor of the NF-kappa-B transcription factor complex. By binding to NF-kappa-B dimers—including p50-p65 (RELA) and p50-c-Rel complexes—it retains them in the cytoplasm, preventing their translocation to the nucleus and subsequent activation of gene transcription critical for immune responses, inflammation, and cell survival. Phosphorylation of NFKBIE targets it for degradation via the ubiquitin-proteasome pathway, thus permitting NF-kappa-B activation in response to various stimuli. NFKBIE is not primarily considered a classic druggable target like a receptor or enzyme, but its regulatory role in immune and inflammatory pathways associates it with disease pathology and therapeutic interest.
