NF-kappa-B inhibitor-interacting Ras-like protein 2 (NKIRAS2) is an atypical Ras-like small GTPase that acts as a potent regulator of NF-kappa-B activity in human cells[6][1][5]. Unlike classical Ras proteins, NKIRAS2 associates with inhibitors of NF-kappa-B (notably NFKBIB/IκBβ) and prevents their signal-induced degradation. This stabilizes NF-kappa-B inhibitors and suppresses the transcriptional activation of NF-kappa-B target genes, thereby modulating inflammatory responses and potentially playing roles in tumorigenesis and other disease states related to chronic NF-kappa-B activation. NKIRAS2 is both nuclear and cytoplasmic and is structurally related to other Ras family GTPases, although it differs functionally by acting as a negative regulator in the NF-kappa-B signaling pathway rather than stimulating proliferation or cell survival[6][1][5].
